2. Cadherin Structure and Homophilic Binding Mechanisms Cadherins possess a conserved structure featuring multiple extracellular cadherin (EC) domains connected by calcium-binding regions, a single transmembrane segment, and a cytoplasmic tail. The calcium-dependent rigidity enables both cis-binding between cadherins on the same cell and trans-binding between adjacent cells. The characteristic knob-and-pocket interaction of EC1 domains ensures homophilic specificity, allowing E-cadherins on epithelial cells to bind exclusively to other E-cadherins. This selectivity is fundamental to tissue sorting during embryonic development and maintains adult tissue boundaries. In pathological conditions like cancer, loss of cadherin expression during epithelial-mesenchymal transition allows tumor cells to escape normal tissue constraints and metastasize to distant sites.

3. Catenin-Mediated Cytoskeletal Linkage Systems Catenins serve as critical adapter proteins connecting cadherins to the cytoskeleton, enabling mechanical strength at cell-cell contacts. L-selectins on lymphocytes enable migration into lymph nodes by binding endothelial ligands. P-selectins on platelets promote aggregation at injury sites, initiating hemostasis and thrombosis. The selectin structure includes an N-terminal lectin domain for carbohydrate binding, an EGF domain regulating binding strength, and variable numbers of SCR domains. 5. Junction Types and Their Specialized Functions Three primary cell-cell junction types serve distinct functions in tissue organization. This is essential in the blood-brain barrier and intestinal epithelium for controlling molecular passage.
