Principles of Chromatography

Retention time is the actual time elapsed from sample injection until the analyte reaches the detector, measured in minutes or seconds. The retention factor (k) is a dimensionless ratio comparing how long the analyte spends in the stationary phase versus the mobile phase. While retention time varies with flow rate and column conditions, the retention factor remains constant under identical chromatographic conditions, making it more useful for method reproducibility and comparison between laboratories.
